What a 5% solution actually contains

A 5% w/v solution contains 5 grams of clascoterone per 100 mL of vehicle. A 1 mL application therefore delivers approximately 50 mg of active to the treated area — a fraction of which crosses the stratum corneum into the follicular unit.

The vehicle is as important as the number. An alcohol-and-glycol carrier system determines penetration, dry-down time and residue, and is the main reason two products with identical concentrations can feel and perform differently.

Why 5% appears so often in the literature

Clascoterone's androgenetic alopecia dose-ranging work compared several concentrations applied twice daily against vehicle. The results were dose-dependent, with the higher arms of the range producing the strongest target-area hair count changes — which is why 5% and above became the concentrations of interest in subsequent development.

Dose response is not linear forever. Beyond a certain point, receptor occupancy saturates and additional active mostly increases the chance of local irritation rather than the size of the effect.

Why 5%?

• 5% w/v — the most widely referenced research concentration; well tolerated in twice-daily use.
• In dose-ranging work, the 5% arm sat at the top of the response curve — higher loads mostly added irritation risk, not effect.
• The total daily quantity of active reaching the scalp matters more than the label percentage alone.
• Scalp tolerability, not concentration, should determine your starting point.

Applying it correctly

Part the hair into sections and dispense along the exposed scalp rather than onto the hair itself. Massage briefly with the fingertips, let it dry fully before styling, and wash your hands.

Avoid applying immediately after microneedling, chemical treatment or any procedure that has compromised the skin barrier — penetration becomes unpredictable and irritation is far more likely.
